Will AA Pro 9 allow me to create a pop-up if I roll my mouse over a word within the text of a paragraph? I would like the users of the PDF to see the definition of a word in a popup when they hold their mouse over that word. Possible? If so, how?

Yes, it's possible. You can create a transparent annotation on top of the word. The contents of the annotation will appear when you hover above it with the mouse.
