Is it possible to highlight AND underline a selection of text using the Comments tools?
 
Trying to mark up a transcript for a Judge highlighting plaintiffs' and defendants' Page and Line Designations and underlining the Objections.

yes, but how you do it depends on whether you have an 'image' (scanned) PDF or a 'text' PDF (created from word processor or the reporter's software). in any case, the most simple way to distinguish plaintiff's statements from defendant's statements would be to use the rectangle tool and assign a different color (say RED to ptf and GREEN to def).

It is text-based, created from e-transcript. I figured out that part. But then I want to underline the Objections that are also highlighted. Is there a way to do both, underline and highlight?

There is not a tool that both highlights and underlines at one time. It would be a two step process. (tip: if you intend to highlight and underline, apply the highlight first and then the underline, particularly if you want access to the tooltip attached to the underline.) I assume that you explored and experimented with all (most of) the 'comment and markup' tools. Right? After you apply the highlight tool to a word or passage, I'd suggest you experiment with the rectangle tool to highlight an objection. I infer that you 'printed' one or more e-Transcript files to PDF. If you're interested in some ideas that you may find useful, I could make available (via Yousendit) an exemplar -- a compendium of 29 transcripts (3600 pp), created from e-Transcript files, which are combined to a single PDF (12 MB). The single PDF is made conveniently useable with comprehensive hyperlinks and bookmarks and made efficiently searchable (all transcripts as a unit) with an embedded index (the index accounts for 4 MB of the 12 MB). Your judge may appreciate having the transcripts, particulary if it's a large set, rendered with the functionaliy of the exemplar.
